1. Four stages of biofilm formation
Biofilms develop in a clear cycle — multiple intervention points exist:
- Stage 1 — Reversible adhesion: scattered bacteria attach to surfaces (pond liner, gut). Easily reversible.
- Stage 2 — Irreversible adhesion: secrete exopolysaccharide (EPS) — detachment becomes hard.
- Stage 3 — Maturation: multi-layer 3D structure, bacteria “communicate” via Quorum Sensing, concentrated toxin output.
- Stage 4 — Dispersion: bacterial clusters break off seeking new surfaces — cycle repeats.
2. Why antibiotics alone don't break biofilms
Thick EPS blocks 70 – 90% of antibiotic diffusion. Core biofilm bacteria switch to “persister” state — slow metabolism, antibiotic-immune. Once antibiotics stop, biofilms re-disperse and disease relapses immediately.
3. Breaking biofilm — Than Vuong's 3-layer strategy
